چکیده

Less than a decade ago the first retroviral oncogene was discovered [1] and shown to derive from a normal cellular gene [2]. To date over 20 different retroviral oncogenes have been found, all derived from cellular counterparts. How a single oncogene can cause cancer is still not fully understood. Natural cancers are known to involve multistep molecular changes, raising a paradox, as compared to cancers induced by retroviral oncogenes. To turn a cellular gene into an active retroviral oncogene, an overexpression due to the very potent retroviral promoter may suffice in a few cases, whereas in most cases several modifications are probably required. These modifications might involve enhanced transcription, truncation of

Insertional Oncogenesis by Non-Acute Retroviruses: Implications for Gene Therapy

Retroviruses cause cancers in a variety of animals and humans. Research on retroviruses has provided important insights into mechanisms of oncogenesis in humans, including the discovery of viral oncogenes and cellular proto-oncogenes. The subject of this review is the mechanisms by which retroviruses that do not carry oncogenes (non-acute retroviruses) cause cancers. The common theme is that th...

Retroviral and human cellular oncogenes.

Unexpected meeting of two separate lines of research resulted in the discovery of oncogenes. Oncogenes are deoxyribonucleic acid (DNA) sequences coding for polypeptide gene products which cause, or contribute to, neoplastic growth of cells. Oncogenes remain almost unchanged through evolution: oncogenes and their gene products of avian, murine, feline, simian and human species show close homolog...

Are activated proto-onc genes cancer genes?

The main objective of cancer molecu~ar biology is to identity cancer genes. ~espite fierce efforts, this objective has still not been met [1-3]. As yet the only known cancer genes are the transformin~ one g~n.e~ of retroviruses. Typically these VIruses InItiate and maintain cancers with autonomous transforming genes that ~re domina~t in susceptible cells [5]. The dIsc~very of ~Ingle gene determ...

Expression of Oncogenes in Human Leukemias1

Oncogenes of retroviruses are directly related to cancers in animals; however, finding relevant associations with human can cer has been difficult. Studies reported here were designed to assess the extent of protooncogene transcription in human leukemias as compared to normal cells. Low-stringency hybridi zation of viral oncogene DNAs to cell messenger RNAs was done to determine whether evoluti...

Murine hematopoietic cells with pre-B or pre-B/myeloid characteristics are generated by in vitro transformation with retroviruses containing fes, ras, abl, and src oncogenes

In vitro infection of bone marrow or fetal liver cells with retroviruses containing fes, abl, ras, or src oncogenes resulted in the transformation of early B lineage cells. All cell lines tested possessed rearrangements at the Ig heavy chain locus and some had rearrangements at the K chain locus. The majority of the lines corresponded phenotypically to Lyb-2+, Ly-5(B220)+, ThB- large pre-B cell...
